The current proposal will characterize two molecular clones of HIV V1-V2 that vary in their susceptibilities to neutralization by V1-specific antibodies. The molecular clones will be compared to their parent viruses using antigenic analysis, pseudotyped virion assays, and cell-cell fusion assays to determine how the resistance to neutralization is conferred. The two molecular clones also contain the V1 loop from a resistant Env from a study that has demonstrated neutralization escape. This work will allow a test of whether the mutation that correlates with neutralization escape is also the one that confers escape from the antiserum. These studies will also determine if the V1-V2 region undergoes gross conformational change during HIV replication. Such conformational change would be visible as an increase in size of the molecular envelope glycoprotein spike, which would be a defining characteristic of these molecular clones of V1-V2. Such structural change would be associated with a change in neutralization sensitivity of Env.
