Binding of 3H-ara-C, 3H-etoposide and 3H-colchicine to these xenografts was compared to binding in multidrug-sensitive xenografts (human colonic adenocarcinoma HT29). We found that the multidrug-resistant xenografts exhibited a lower binding of 3H-ara-C (0.4 +\/- 0.2 versus 1.1 +\/- 0.2 pmol\/mg protein) and 3H-etoposide (0.7 +\/- 0.1 versus 1.7 +\/- 0.3 pmol\/mg protein) but a higher binding of 3H-colchicine (1.3 +\/- 0.1 versus 0.8 +\/- 0.1 pmol\/mg protein) compared to that of the multidrug-sensitive xenografts. Similar differences were found in the mouse tissues as determined by liquid scintillation counting and radioreceptor assay (RRA). The lower binding of 3H-ara-C in multidrug-resistant tumors could not be explained by differences in nuclear localization or mRNA levels of the multidrug-resistant protein compared to the multidrug-sensitive xenografts. The binding of 3H-etoposide and 3H-colchicine, however, appeared to be dependent on the level of mRNA and protein of P-glycoprotein as determined by cDNA hybridization analysis and immunoblotting, respectively. The higher specific binding of 3H-colchicine and 3H-etoposide in xenografts compared to mouse tissues was indicated by the high levels of specific binding in multidrug-resistant xenografts.
