Working with Photoshop: A Quick Tour In this chapter, I take you through the most common features in Photoshop to help you get off to a good start. Detection of advanced prostate cancer is dependent on molecular markers. Prostate cancer is the second leading cancer amongst men in the United States, with 250,000 new cases estimated in the year 2000. Due to a lack of initial and subsequent recurrences, surveillance of advanced prostate cancer is not routinely done. The discovery of aggressive prostate cancers may be aided by molecular markers that correlate with disease progression. We evaluated the expression of CK5/6, Ki-67, p63, androgen receptor (AR), and PTEN by immunohistochemistry and their utility in detecting advanced prostate cancer. Serum prostate-specific antigen (PSA) was also evaluated as a diagnostic tool. Cases were selected from the archives of the Department of Pathology at the University of Nebraska Medical Center between 1992 and 2000, based on the presence of lymph node metastasis or a positive surgical margin (PSM) after radical prostatectomy. Cases were categorized into two groups based on patient follow-up, from 1 to 4 years or > or =5 years. Average follow-up was 19.8 months (range 7-32). A total of 146 cases were evaluated, including 101 cases with lymph node metastasis and 45 with PSM. The average age of cases was 64.5 years (range 42-83). All PSA values were in the range 1-134 ng/mL (mean 31 ng/mL). PSA values increased with the stage of disease. Expression of Ki-67 was evident in 26 of 101 cases of lymph node metastasis and 6 of 45 cases with PSM. Expression of CK5/6 was seen in 34 of 101 cases with lymph node metastasis and 9 of 45 cases with PSM. Expression of AR was evident in 74 of 101 cases with lymph node metastasis and 16 of 45 cases with PSM.
